Effect of iba concentration on in vitro root regeneration of different mint explants

An experiment was conducted in the Tissue Culture Laboratory of the Department of Horticulture, Bangabandhu Sheikh Mujibur Rahman Agricultural University, Gazipur 1706, from March 2013 to February 2014 for in vitro root regeneration of mint {Mentha sp.) using different explants and IBA in full strength MS media. Three different explants (viz., nodal segment, shoot tip and leaf) were evaluated using three levels of IBA (1.0,
2.0, and 3.0 mg/L) along with control for root regeneration. Results revealed that shoot
tip and nodal segment performed better than leaf as explant in all the studied characters.
Shoot tip and nodal segment initiated root within the shortest time of 13.8 and 14.6 days, respectively, with 1.0 mg/L of IBA; while in leaf, it took the shortest time of 34.8 days with 2.0 mg/L of IBA. Statistically similar results in number of roots per explant and length of the longest root were recorded in shoot tip and nodal segment along with 1.0 mg/L of IBA in almost all days after inoculation. Leaf showed the poorest performance regarding root regeneration. Therefore, for in vitro root regeneration of mint 1.0 mg/L of IBA may be used with shoot tip or nodal segment explant.
